What Is Lapachone?
Lapachone is a naphthoquinone, a type of organic compound extracted from the bark of the lapacho tree. It exists in two main forms: α-lapachone and β-lapachone. In skincare, it is primarily valued for properties that may support the skin's natural repair processes.
The compound has been traditionally associated with various biological activities, and contemporary research has begun to investigate its potential application in formulations designed to support wound healing and skin recovery.
How It Works
Lapachone appears to support skin repair through multiple mechanisms. Research suggests it may upregulate Sirt3, a protein involved in cellular energy regulation and stress response. This activation may encourage migration of skin cells and promote angiogenesis — the formation of new blood vessels — which is essential for delivering oxygen and nutrients to healing tissue.
The compound's quinone structure gives it antioxidant and antimicrobial properties, which may help create an environment conducive to faster tissue regeneration. However, the precise mechanisms in human skin remain incompletely understood, and most evidence comes from in vitro and animal studies.
Clinical Evidence & References
Pre-clinical research has shown promising results. Mouse studies demonstrated that β-lapachone treated burn wounds recovered approximately 30% faster than untreated controls by day three, with histological evidence of accelerated tissue regeneration across multiple skin layers.
The research base is still developing. Current evidence is primarily derived from animal models and laboratory studies rather than human clinical trials. Whilst these early findings are encouraging, larger, well-controlled human studies are needed to confirm efficacy and establish optimal concentrations for topical use. The ingredient remains relatively understudied compared to more established skincare compounds.
How to Use
Lapachone is typically incorporated into creams, serums or ointments designed to support healing. As a supporting ingredient rather than a standalone active, it works best as part of a formulation that includes other evidence-backed repair ingredients. Use as directed by the product instructions, and if applying to compromised or broken skin, ensure the formulation is appropriate for that use case.
