What Is Estradiol?
Estradiol (17β-estradiol) is a form of oestrogen, a hormone naturally produced in the body. In skincare, it functions as a skin conditioning agent. It is sometimes derived from plant sources such as soy or yam, though synthetic versions also exist.
The ingredient is included in skincare formulations to help maintain skin health and resilience, particularly in relation to age-related changes in the skin's appearance and function.
How It Works
Estradiol appears to work through multiple pathways. At the cellular level, it has antioxidant properties, helping to neutralise reactive oxygen species that accumulate with age and environmental exposure. These ROS molecules can damage proteins, lipids and DNA, contributing to visible signs of ageing.
The hormone also interacts with oestrogen receptors in skin cells, signalling pathways that regulate skin thickness, vascularisation and the production of structural proteins like collagen. This is thought to be why oestrogen levels have a noticeable effect on skin health during hormonal transitions.
Clinical Evidence & References
In-vitro studies show that estradiol can protect human skin fibroblasts and keratinocytes against oxidative damage when exposed to hydrogen peroxide, suggesting a protective mechanism at the cellular level. Research also indicates that oestrogen contributes to skin thickness, vascularisation and collagen maintenance — changes particularly noticeable during perimenopause when oestrogen levels decline.
However, the relationship between topical estradiol and systemic oestrogen is complex, and evidence for the efficacy of topically applied estradiol in cosmetic products remains limited. Larger clinical trials would be needed to establish clear benefits in finished skincare formulations at typical concentrations.
How to Use
If using a product containing estradiol, apply as directed on the product label, typically once or twice daily to clean skin. The ingredient is often included in moisturisers or serums intended for mature skin. Consistency over time is more relevant than frequency when using skincare actives.
