What Is Tetrapeptide-68?

Tetrapeptide-68 is a short-chain peptide derived from Loricrin, a structural protein found naturally in the skin. Peptides are fragments of proteins composed of amino acids linked together, and those with 2-50 amino acids are increasingly used in skincare for their stability and efficacy.

In formulations, Tetrapeptide-68 functions as a skin conditioning agent. The ingredient is thought to work by communicating with skin cells to support the maintenance of skin structure and texture, potentially influencing the appearance of fine lines and surface roughness.

How It Works

Tetrapeptide-68 is believed to communicate with skin cells through receptor-mediated pathways. When applied topically, it may signal fibroblasts and other skin cells to support the maintenance of collagen and elastin networks that naturally decline with age.

By reinforcing these structural proteins, the peptide can help improve skin firmness and reduce the depth of fine lines. This mechanism is gentler than more aggressive anti-ageing approaches and typically causes minimal irritation.

Clinical Evidence & References

A 12-week randomised controlled trial in 25 women aged 30-65 years found that a formulation containing Tetrapeptide-68 at 100 ppm produced significant improvements in periorbital wrinkles and skin roughness parameters, measured both instrumentally and via 3D imaging. Participants also reported positive efficacy and usability assessments.

Whilst these results are encouraging, the study was relatively small. Additional research with larger cohorts and longer follow-up periods would help establish the ingredient's efficacy more firmly, particularly for different skin types and concerns beyond periorbital wrinkles.

How to Use

Tetrapeptide-68 is typically found in eye creams and lightweight moisturisers intended for daily use. Apply a small amount around the orbital bone, avoiding direct contact with the eye itself. Peptides work best when used consistently over several weeks, so regular application is recommended for visible results. They pair well with other gentle actives like hyaluronic acid and niacinamide.

Safety & Precautions

Tetrapeptide-68 is generally well-tolerated with a low irritation profile typical of peptide ingredients. It is suitable for most skin types, including sensitive skin, though any new product should be patch-tested first.
