What Is Glycolide?

Glycolide is a monomer — a small molecular unit — that forms the basis of poly(lactide-co-glycolide), commonly known as PLGA. This is a synthetic, biodegradable polymer derived from aliphatic polyesters. In skincare formulation, glycolide itself is not typically used as a standalone ingredient; rather, it exists as part of PLGA copolymers employed in specialised drug delivery systems.

The primary function of PLGA-based formulations in skincare is to act as a controlled-release vehicle for active ingredients. These polymers can encapsulate drugs or actives and release them gradually over time, allowing for sustained efficacy and potentially improved skin penetration.

How It Works

Glycolide, as part of a PLGA copolymer, functions as a structural component of microparticles or thin films. When formulated correctly, these polymeric structures create a skin or shell that forms rapidly — sometimes within milliseconds — around encapsulated active ingredients. This barrier controls the rate at which the active is released into the skin.

The polymer's degradation occurs through hydrolysis, breaking down into lactic and glycolic acid byproducts that the body can process naturally. This mechanism allows skincare actives to be delivered more steadily than they would be in conventional formulations, potentially improving their efficacy.

Research suggests that PLGA thin films support normal skin cell behaviour in vitro. Studies have shown that human keratinocytes and fibroblasts cultured on PLGA films maintain normal morphology, focal contacts, cytoskeleton organisation, and differentiation patterns compared to control conditions. Proliferation and migration parameters also remained within normal ranges.

However, evidence for PLGA-based skincare benefits in human clinical trials is limited. Most research to date has focused on the polymer's use in pharmaceutical and tissue engineering applications rather than cosmetic efficacy. Further in vivo studies would be needed to establish concrete skincare benefits for end users.

How to Use

If you are using a skincare product containing PLGA delivery systems, apply according to the product instructions. These formulations are designed to work over time, so consistent use is more important than frequency. Allow the product to absorb fully before applying other treatments, as the controlled-release mechanism works best when the product remains undisturbed on the skin.

PLGA is generally well-tolerated and has a long history of safe use in pharmaceutical and medical device applications. As a biodegradable synthetic polymer, it poses minimal risk of accumulation.
