What Is MATRIXYL 3000?

MATRIXYL 3000 is a trade-name peptide complex used in anti-ageing skincare. On an ingredients list it usually appears as its two active components rather than under the trade name: Palmitoyl Tripeptide-1 and Palmitoyl Tetrapeptide-7, sitting alongside the glycerin, water and butylene glycol carrier the complex is supplied in.

Both are peptides — very short chains of amino acids, the same building blocks that make up structural proteins such as collagen and elastin. Palmitoyl Tripeptide-1 is a three-amino-acid sequence (glycine, histidine, lysine). Palmitoyl Tetrapeptide-7 is a four-amino-acid sequence (glycine, glutamine, proline, arginine). Each is bonded to palmitic acid, a fatty acid, which makes the peptide more oil-soluble so it spreads through the upper layers of the skin's surface rather than simply sitting on top of it.

Both peptides are made synthetically in a laboratory rather than extracted from plants or animals, so the sequence and purity stay consistent from batch to batch. The peptides themselves are used at very low levels, and that is by design: the trade complex is added at a small percentage of the finished formula, so the individual peptides are present only in trace amounts.

You will find the complex in serums, eye products and moisturisers aimed at the appearance of fine lines, crepey texture and skin that has lost some of its bounce. It is usually one of several actives in a formula rather than the sole point of it.

How It Works

Peptides like these are described as signal or messenger peptides. The thinking behind them rests on matrikines: fragments released when the skin's own matrix proteins break down, which cells appear to read as instructions.

Palmitoyl Tripeptide-1 corresponds to a fragment of the collagen molecule. In laboratory work on isolated cells, this fragment has been used to study how skin cells respond to collagen breakdown products. Applying the peptide is intended to mimic that prompt without any breakdown having actually occurred.

Palmitoyl Tetrapeptide-7 has been studied on a separate pathway. Laboratory work on isolated cells suggests it influences signalling molecules involved in the turnover of matrix proteins. This is test-tube observation only and says nothing about what happens on skin in use.

The rationale for pairing the two is that they were selected for different laboratory endpoints rather than the same one. Whether that pairing translates into a visible difference on skin is not well established outside the supplier's own testing.

Be realistic about the evidence. Much of the published support comes from the supplier's own testing, independent human trials are limited, and any change is gradual and modest in appearance rather than dramatic.

How to Use

Peptide complexes are low-drama actives. Consistency and sensible layering matter far more than dose-chasing.

  • Apply to clean, slightly damp skin. Peptide serums are usually water-based, so they belong early — after cleansing and any watery toner, before richer creams and oils.
  • Use it twice a day and judge it over about two months. The supplier's own testing used the complex at 3%, applied morning and evening for roughly eight weeks, so match that and assess at the end, not after a fortnight.
  • Separate it from very low-pH layers. If you also use a direct acid or a high-strength vitamin C, apply them at a different time of day rather than stacking them wet-on-wet.
  • It layers happily with most things. Niacinamide, hyaluronic acid and ceramides are easy partners, and peptides sit well alongside retinoids — apply the peptide serum first, then the retinoid.
  • Always follow with SPF in the morning. Daily sun protection does more for the look of ageing skin than any peptide.
  • Store it sensibly. Opaque, air-restricted packaging keeps a formula in better condition than an open jar.

Safety & Precautions

These peptides are among the better-tolerated cosmetic actives. They do not sting or tingle the way acids can, and there is no known photosensitivity associated with them — though daily sunscreen remains sensible regardless.

They generally suit oily, dry, combination and sensitive skin. As with any cosmetic ingredient, an allergic reaction is possible, so patch test a new product on the inner forearm for a few days first. Reactions to peptide serums are more often traced to fragrance or other formula components than to the peptides themselves.
