What Is Nicotinoyl Dipeptide-22?
Nicotinoyl Dipeptide-22 is a synthetic peptide derived from niacinamide, a well-established skincare ingredient. It belongs to a class of niacinamide derivatives engineered to target skin pigmentation through a different mechanism than traditional tyrosinase inhibitors.
The ingredient works primarily as a skin conditioning agent, with research suggesting it may help reduce the appearance of uneven tone and pigmented spots when incorporated into topical formulations.
How It Works
Rather than directly inhibiting tyrosinase (the enzyme that produces melanin), Nicotinoyl Dipeptide-22 appears to work by suppressing the transfer of melanosomes — the pigment-containing structures within melanocytes — to surrounding skin cells. This indirect mechanism may help reduce visible pigmentation without disrupting normal cellular function.
The ingredient also retains antioxidant properties, which may offer additional protective benefits to the skin barrier.
Clinical Evidence & References
In laboratory and reconstructed skin models, niacinamide derivatives in this class have demonstrated the ability to reduce melanin production and lighten skin appearance at concentrations around 0.05–0.1%. A small human trial (52 volunteers) using a 0.1% formulation showed no signs of irritation or sensitisation.
However, it is important to note that most available evidence relates to similar niacinamide derivatives rather than Nicotinoyl Dipeptide-22 specifically. Whilst results are promising, larger clinical studies would help establish efficacy and optimal concentration ranges for this particular ingredient.

How to Use
Nicotinoyl Dipeptide-22 is typically found in serums, essences, and targeted treatment products. Apply to cleansed skin as directed by the product formulation. It can generally be incorporated into morning and evening routines, though patch testing is advisable if you have sensitive skin or are using multiple active ingredients.
